Using eDNA Metabarcoding to Monitor Changes in Fish Community Composition After Barrier Removal

Artificial instream barriers are a major cause of habitat fragmentation that reduce population connectivity and gene flow by limiting fish movements. To mitigate their impacts, obsolete increasingly been removed worldwide, but few barrier removal projects monitored. We employed powerful Before-After-Downstream-Upstream (BADU) approach using environmental DNA (eDNA) metabarcoding to examine the effects on community composition removing weir in river Lugg (England) had suggested have detrimental effect salmonid migration. found no change diversity or relative abundance after above below weir, detected an important sampling season, likely related species' life cycles. eDNA nine species were also identified electrofishing one additional ( Anguilla anguilla ) was missed traditional surveys. Our results suggest monitoring should be carried out ensure any ecological benefits properly documented is sensitive technique monitor removal.
